Recognizes a protein of 75 kDa, identified as γ heavy chain of human immunoglobulins. It does not cross-react with α (IgA), μ (IgM), ε (IgE), or δ (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Biotium CD66 (CEA)(C66/195), CF568 conjugate, 0.1mg/mL
This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb reacts with nonspecific cross-reacting antigen (NCA) and shows a cross-reaction with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Podocalyxin is a member of the CD34 transmembrane sialomucin family. It is highly on kidney podocyte foot projections and plays essential roles in kidney development and homeostasis, blood filtration and urine formation. It is also expressed on vascular endothelia, hematopoietic progenitors and a subset of neurons. Overexpression of podocalyxin may be linked to more aggressive tumor behavior. This antibody recognizes a protein of 25-26 kDa, identified as Bcl-2. This antibody shows no cross-reaction with Bcl-x or Bax proteins. Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. Bcl-2 appears to function in a feedback loop system with caspases. It inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1). Expression of Bcl-2 inhibits programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of Bcl-2 protein, whereas the normal or hyperplastic germinal centers are negative. Recognizes Nucleolin (NCL), the major nucleolar phosphoprotein of growing eukaryotic cells. NCL is located mainly in dense fibrillar regions of the nucleolus. It is found associated with intranucleolar chromatin and pre-ribosomal particles. Human NCL gene consists of 14 exons with 13 introns and spans approximately 11 kb. It has a predicted molecular weight of ~76 kDa with an apparent MW of 100-110 kDa, attributed to phosphorylation of its N-terminal domain. It induces chromatin decondensation by binding to histone H1. It is thought to play a role in pre-rRNA transcription and ribosome assembly.This MAb can be used to stain the nucleoli in cell or tissue preparations and can be used as a marker of the nucleoli in subcellular fractions. Recognizes a protein of a disulfide-linked 120 kDa dimer, identified as CD27 (Workshop VI; Code 6T-028). CD27 is expressed on the majority of peripheral T cells, medullary thymocytes, memory-type B cells, and natural killer cells. It is a transmembrane phosphoglycoprotein that belongs to the tumor necrosis factor receptor (TNFR) superfamily. CD27 binds to its ligand CD70, a member of the TNF family, and induces T-cell co-stimulation and B-cell activation. CD46 acts as a cofactor for complement factor I, a serine protease, which protects autologous cells against complement-mediated injury by cleaving C3b and C4b deposited on host tissue. It may be involved in the fusion of the spermatozoa with the oocyte during fertilization. CD46 acts as a co-stimulatory factor for T-cells, which induces the differentiation of CD4 into T-regulatory 1 cells.
